Identification of the causal agent of rice black – streaked dwarf disease in the North of Vietnam in 2009

In summer crop, 2009, a serious drwarf disease of rice occurred in several provinces in the North. The disease has symptoms similar to those of the rice ragged stunt disease (rice ragged stunt virus, RRSV) in the South like stunt, twisted leaves, dark leaves. However, the distinct symptoms like small white to brown or black waxy galls along veins of culms, particulary near base, and the lack of yellow ragged appearance in one edge of the tweasted or recovering leaves suggested the disease in the North may be caused by a rreovirus like Rice black – streaked dwarf virus (RBSDV) and the related virus, namely Southern Rice black – streaked dwarf virus (RBSDV2). The universal primers specific for both RBSDV and SRBSDV were designed based on conserved regions of the genomic S10 fragment. RT – PCR test on typical samples collected from different provinces gave products with expected sizes using this primers but were negative for primers specific RRSV. Four RT – PCR products representative for 4 provinces Nghe An, Thanh Hoa, Nam Dinh and Son La were directly sequenced. Blast search and sequence analyses showed they were all isolates of SRBSDV. The virus was taken photo with electron microscope JEOL 1010.
